Extracellular matrix requirements for gastrointestinal organoid cultures

چکیده
Organoids are a new class of biological model systems that have garnered significant interest in the life sciences. When provided with proper 3D matrix and biochemical factors, stem cells can self-organize form tissue-specific organoids. Thus far, there has been substantial effort to identify soluble niche components essential for organoid culture; however, role solid extracellular (ECM) as an element is still largely lacking. In this review, we discuss importance ECM intestinal, hepatic, pancreatic culture how biomaterial-based approaches be used probe different properties required more physiologically translationally relevant models.

Extracellular Matrix Bioscaffolds for Building Gastrointestinal Tissue
Regenerative medicine is a rapidly advancing field that uses principles of tissue engineering, developmental biology, stem cell biology, immunology, and bioengineering to reconstruct diseased or damaged tissues. Biologic scaffolds composed of extracellular matrix have shown great promise as an inductive substrate to facilitate the constructive remodeling of gastrointestinal (GI) tissue damaged ...
متن کاملCultures of human retinal pigment epithelium. Modulation of extracellular matrix.
The extracellular matrix of retinal pigment epithelial (RPE) cells is an important component of the fibroelastic membranes formed in proliferative vitreoretinopathy (PVR). These membranes consist of cells intermingled with fibrillar proteins, such as elastin and collagen. Because RPE cells may be exposed to vitreous and serum-derived factors in conditions that lead to PVR, the effects of vitreo...
متن کاملOrganoid Cultures Derived from Patients with Advanced Prostate Cancer
The lack of in vitro prostate cancer models that recapitulate the diversity of human prostate cancer has hampered progress in understanding disease pathogenesis and therapy response. Using a 3D organoid system, we report success in long-term culture of prostate cancer from biopsy specimens and circulating tumor cells. The first seven fully characterized organoid lines recapitulate the molecular...
متن کاملMatrix Rigidity-Modulated Cardiovascular Organoid Formation from Embryoid Bodies
Stem cell clusters, such as embryoid bodies (EBs) derived from embryonic stem cells, are extensively studied for creation of multicellular clusters and complex functional tissues. It is common to control phenotypes of ES cells with varying molecular compounds; however, there is still a need to improve the controllability of cell differentiation, and thus, the quality of created tissue. This stu...
متن کاملRegulatory and Safety Requirements for Food Cultures
The increased use of food cultures to ferment perishable raw materials has potentiated the need for regulations to assess and assure the safety of food cultures and their uses. These regulations differ from country to country, all aimed at assuring the safe use of food cultures which has to be guaranteed by the food culture supplier. Here we highlight national differences in regulations and rev...
